Today, we know black holes exist, and now scientists are trying to confirm that other holes lurk in hyperspace. Our infinite cosmos could contain a variety of "holes" such as black, white, "mini" and wormholes. White holes are the reverse of black holes; instead of matter being sucked into it, matter is ejected out. Wormholes are gateways in the fabric of space and time. They are included in Einstein's field equations as possibilities for their existence. Neither white holes nor wormholes have ever been found. The cycle of life of a handful of different animals is captured on film in a whole new way in this documentary. Using special motion-control photography equipment, Genesis allows filmgoers to view animal behaviors which are too small, too slow, or too difficult to normally be seen with the naked eye, including a chick hatching its way out from inside an egg, jellyfish drying into nothing under the heat of the sun, or a snake slowly swallowing prey bigger than itself. This footage is accompanied by narration from Sotigui Kouyate, who uses simple props and easily understandable analogies to explain the science behind what its shown on screen. Genesis was directed by Claude Nuridsany and Marie Perennou, who previously created another acclaimed scientific documentary, Microcosmos.

Thirty years ago, scientists first realized that some unknown dark substance was affecting the way galaxies moved. Today, they think there must be five times as much dark matter as regular matter out there. But they have no idea what it is - onl that it's not made of atoms, or any other matter we are familiar with. The ship journeys through the universe's hundred billion galaxies, the Local Group, the Andromeda Galaxy, the Milky Way, the Orion Nebula, our Solar System, and finally the planet Earth. Eratosthenes' successful calculation of the circumference of Earth leads to a description of the ancient Library of Alexandria. Finally, the Ages of Science are described, before pulling back to the full span of the Cosmic Calendar.

Sagan discusses the story of the Heike crab and artificial selection of crabs resembling samurai warriors, as an opening into a larger discussion of evolution through natural selection (and the pitfalls of intelligent design). Among the topics are the development of life on the Cosmic Calendar and the Cambrian explosion; the function of DNA in growth; genetic replication, repairs, and mutation; the common biochemistry of terrestrial organisms; the creation of the molecules of life in the Miller-Urey experiment; and speculation on alien life (such as life in Jupiter's clouds). In the Cosmos Update ten years later, Sagan remarks on RNA also controlling chemical reactions and reproducing itself and the different roles of comets (potentially carrying organic molecules or causing the Cretaceous–Paleogene extinction event).
